Psoriasis biologics linked to lower depression and anxiety scores

A network meta-analysis finds every biologic class beat placebo on mood as well as skin; alongside, risankizumab against ustekinumab in children, a dual JAK1/TYK2 inhibitor in eczema, and three-year facial data for lebrikizumab.

The edition in brief

A BJD network meta-analysis of eight studies (4,319 adults with moderate-to-severe psoriasis) found every biologic class was associated with improved depression and anxiety symptoms against placebo; IL-12/23 inhibitors had the largest depression estimate (SMD 0.68, 95% CI 0.47 to 0.88). Mood generally improved alongside skin, though not in strict proportion, and drug rankings were uncertain. The phase 3 OptIMMize-1 study in 137 children with moderate-to-severe psoriasis found risankizumab and ustekinumab gave similar week-16 clear or almost-clear rates (79.6% vs 75.0%), with complete clearance more frequent on risankizumab (40.7% vs 17.9%) and responses held to week 52. A phase 2 trial of QY201, an oral JAK1/TYK2 inhibitor, gave EASI-75 in 72.9% to 84.7% of adults with atopic dermatitis against 18.5% on placebo at 12 weeks. A post hoc analysis of lebrikizumab trials showed 94.8% of week-16 responders kept head-and-neck EASI-75 at three years. The pearl covers comorbidity screening in psoriasis.
