After surgery for bone and joint infection with local antibiotics, a week of systemic therapy was enough

A 500-patient trial found seven days no worse than four weeks or more, with far fewer side effects. Also: a quarter of body weight lost on retatrutide, a negative phase 3 trial of faecal transplant for IBS, and an oral on-demand drug for hereditary angioedema.

The edition in brief

The SOLARIO trial randomised 500 adults who had surgery for orthopaedic infection with an implanted local-antibiotic carrier to seven days or less versus four weeks or more of systemic antibiotics. Treatment failure at 12 months was 11.1% with the short course and 14.1% with the long, meeting non-inferiority, and treatment-related symptoms by week six fell from 45.2% to 17.2%. TRIUMPH-1, a phase 3 trial of 2,339 adults with obesity without diabetes, found once-weekly retatrutide, which acts on GIP, GLP-1 and glucagon receptors, reduced body weight by 23.7% and 25.0% at the two higher doses over 80 weeks, against 3.9% with placebo, and also reduced knee osteoarthritis pain and sleep apnoea events. REFIT2, a Norwegian phase 3 trial in 448 adults with moderate to severe irritable bowel syndrome, found a single donor faecal transplant no better than the patient's own faeces at 90 days. RAPIDe-3, a crossover trial in 134 people with hereditary angioedema, found oral deucrictibant brought symptom relief at a median of 1.28 hours against more than 12 hours with placebo.
