FDA approves a supplement to the pirtobrutinib application

Note the pirtobrutinib label update and read the revised label before assuming any change in indication or dosing.

On 2 October 2026 the FDA approved a supplement to the new drug application for pirtobrutinib (Jaypirca), held by Loxo Oncology. The notice does not state what the supplement changes; supplements can cover labelling, dosing, manufacturing or new indications.

Pirtobrutinib is a non-covalent BTK inhibitor used in B-cell malignancies. Until the label change is published, prescribers should not assume any new indication or dosing advice. Its status with CDSCO is not stated in this notice.

  • The notice confirms an approved supplement but not its content.
  • Check the updated FDA label before changing practice.
  • As with other BTK inhibitors, review interacting CYP3A drugs and bleeding risk with anticoagulants or antiplatelets.
  • Indian regulatory status is not addressed by this notice.

Why it matters

BTK inhibitor labels carry interaction and bleeding advice that clinical pharmacologists are asked about.

Don't overread it

A licence supplement is not, on its own, a new indication.
